The Promist™ delivery system is a breakthrough in pet medicines. It provides several significant advantages when compared to conventional swallow tablets and liquids. These advantages, either individually or in combination, can make a considerable contribution to developing a superior veterinary pharmaceutical. These advantages include:
- Convenient dosing leading to increased compliance
- Improved bioavailability by avoiding the first-pass liver effect
- Faster absorption and onset of therapeutic activity
- Extended product life cycles via patented novel delivery system
For years, pet owners have struggled with dosing pets with tablets. Since swallowing a pill is unnatural for pets, they often resist by attempting to escape, squirm, or in some cases, bite. Frustrated owners often resort to tricks, games, wrestling and even brute force. In addition, even if the owner gets the pet to take the pill, the animal may spit it out or regurgitate it. If and when this occurs beyond the sight of the owner, the pet goes untreated and the health benefits of the medicine are lost. With Velcera's oral mucosa delivery system, a precise metered dose of the medication can be sprayed as a fine mist on the inside surface of the animal's cheek and gums. This process is significantly easier on both the pet and the owner. With no volume of medicine for the animal to swallow or spit out, the owner can be confident the pet has been treated; thereby increasing compliance with the veterinary-recommended treatment.

The liver serves as a filter for metabolizing compounds in the body. The liver may break down an active pharmaceutical ingredient injested via the gastrointestinal system before it has a chance to deliver its therapeutic effect. This is referred to as the 'first-pass liver effect'. In drugs with a high first-pass liver effect, increased drug dosage needs to be given in order to deliver the desired therapeutic effect.
By avoiding the gastrointestinal system and the first-pass liver effect, Promist can deliver the same therapeutic benefit with less active ingredient. This may make a previously unusable drug useful. Reducing the required amount of active ingredient can also lower the cost of manufacture of the medication.
Promist offers potential lower dosing for the same therapeutic effect.
Figure 1:
A comparison of absorption stages of Promist versus oral formulations
Figure 2: A comparison of plasma concentrations of active pharmaceutical ingredient using Promist Vs. Tablet, Canine, Compound VEL-501
With the drug presented in Figure 2, peak plasma concentrations are much higher and occur much sooner than with a tablet form. In this case, Promist delivery increases the bioavailability of the active ingredient, and should be able to improve performance.
Since a medication using PromistT is absorbed quickly and directly through the mucosal surface into the bloodstream, the active ingredient may be able to go to work significantly faster than traditional tablet and liquid formulations.
Since there is no pill or liquid to swallow, Promist has the added potential benefit of reducing the potential for gastrointestinal side effects or adverse food reactions.
With some drugs, plasma concentrations are very similar to the traditional form, but with faster absorption. In these cases, we can expect Promist delivery to result in the same or equivalent performance as a traditional form, but with greater convenience and faster onset of action. Promist is a patented drug delivery technology, which can extend the patent life of any medication that is reformulated based upon this proprietary delivery system. This can bring new value to strategic partners, extending the profitable life of their commercialized products and brands.
